DSIP Reconstitution and Spray Calculation for a 5 mg Vial (5 mL Nasal Spray Bottle)

Materials
- 10 mg DSIP vial (lyophilized powder)
- 5 mL bacteriostatic water
- 10 mL sterile nasal spray bottle (0.1 mL/spray pump)
- 1 mL insulin syringe
Reconstitution and Preparation Steps
- Draw Water: Using a 1 mL insulin syringe, draw 1 mL bacteriostatic water (100 units).
- Reconstitute DSIP at Home: Inject 1 mL bacteriostatic water slowly into the 10 mg DSIP vial along the side. Gently swirl until fully dissolved (clear solution; discard if cloudy). Why reconstitute at home? DSIP is a peptide, and excessive shaking or agitation (e.g., during transport) can denature or degrade its protein structure, potentially reducing its efficacy. Reconstituting at home minimizes handling and ensures gentle mixing to preserve the peptide’s integrity.
- Transfer to Spray Bottle: Draw the 1 mL DSIP solution into the syringe. Inject into a 10 mL sterile nasal spray bottle. Add 4 mL more bacteriostatic water to the spray bottle to reach a total volume of 5 mL (final concentration: 2,000 mcg/mL).
- Prime Spray Bottle: Secure the nozzle and spray 3–5 times into the air until a fine mist forms.
- Label: Label the bottle with the date, concentration (2,000 mcg/mL)
Storage Guidelines
- Unreconstituted (Powdered Vial): Store at room temperature (cool, dry, dark) for up to 2 months or freeze at -20°C for 6–12 months.
- Reconstituted Solution (in Spray Bottle): Refrigerate at 2–8°C; use within 60 days. Do not freeze. Protect from light.
Spray Calculation
- Total Volume: 5ml
- Spray Output: 0.1 mL/spray (standard metered pump)
- Total Sprays: 5mL ÷ 0.1 mL/spray = 50 sprays
Dosage per Spray
- Total DSIP: 10 mg = 10,000 mcg
- Total Volume: 5 mL
- Concentration: 10,000 mcg ÷ 5mL = 2,000 mcg/mL
- Dosage per Spray: 0.1 mL × 2,000 mcg/mL = 200 mcg DSIP per spray
